Boone Carlyle and Shannon Rutherford (the brother and sister) were killed off in accidents. Edward Mars (the marshal) also died after the crash when Jack euthanized him.
Elizabeth Rutherford ("Libby") and Ana Lucia Cortez were also killed off when they were shot by Michael, but they were newer introductions to the cast. Dr. Arzt (a newer arrival) blew himself to pieces with dynamite. Goodwin (an Other) was killed by Ana Lucia - wait, he was a newer arrival too.
Michael and Walt looked like they were leaving the show at the end of season 2, but I don't think we've seen the end of them yet.
So, I think that makes 3 of the original cast that have left the show, and a bunch of other cast members that were introduced later on.

To the best of my knowledge, we're down two people. The killed off Boone and Shannon in separate accidents. I had forgotten about the Marshall, Sawyer shot him, but not fatally.
Libby and Ana Lucia were killed off, too. However their characters weren't introduced until Season 2
