Go to My computer, right click, then select properties... Choose Device Manager, and look under soundcards, media, etc... See if there are little yellow explanation points beside anything. If there are, search for drivers specific to the hardware online, download them. Then, in device manger, uninstall the device, then search for new hardware. When it picks it up, it'll ask for new drivers. Either install the new drivers, or guide the wizard to the location in which you saved them.
But before you do all of this, make sure mute isn't selected in the volume controls, and that the plug is plugged into the proper place.
Also, to find out which hardware you're using, the name of the card is in Device Manager. Get the model number and all, then look for the drivers.

you may need to reinstall your audio driver. Try this go to start menu. control panel.
Select sounds speach and Audio Devices. then select that again.
Check speaker volume on this panel, also check for any error symbols.
Click on hardward and then properties. uninstall/reinstal button

What kind of computer ?
What operating system ?
Did you do the sound recording test at; Start / Programs / Accessories / Entertainment / Sound Recorder ?
Did you try headphones to rule out the speakers ?
Are you in the Mute mode ?
