To make sure the readership is right for your vacancy you should have a copy of the publication first. There will be publishers details somewhere, and you want to contact whoever is in charge of 'sales' (as opposed to editorial). This info is usually located towards the front of the newspaper/magazine.

Contact the publishers, I work for a magazine company and each publisher does several titles and they will have only 1 advertising department for everything they publish.
It depends on what you want to advertise as to who you contact as some publishers only do certain types of magazines.
