Depends.
Who is the assailant? How big? Does the assailant know Martial Arts? Is the assailant carrying any form of weapon?
Is the .45 handgun loaded? Is it ****** and ready to fire, or I will have to pull the slide back and chamber a round? Do I have time to chamber or a round? Is a round already chambered?
How experienced am I in Karate? Am I a Black Belt?
Answers to these questions would be better in order to give a proper response.
However, being a Tang So Do Karate Orange belt, and after witnessing my Masters skills, I would choose Karate. Much funner to subdue and stomp on someone with your own bare hands than having to use a tool.

Depends on how big they are and how many of them. If one short guy use Karate. Anything other then that use a 45! But if he is an ex NFL linemen use a 44!
