They don't test for nicotine. Only for illegal or illicit drugs.
Unless not smoking is a requirement for your job, in which case they just might. Nicotine does not stay in the body long, that's why smokers need to smoke so frequently, to replenish the depleted drug. A couple of days should remove all traces.

Standard drug tests don't test for nicotine. But it can be tested for if its asked for specifically. Different drugs stay in your system for different times. Like cocaine, will stay in your urine for up to 3 days AT MOST, despite various factors. With something like marijuana or nicotine, it depends on everything from the metabolism of the person, how often one smokes, the strength of whats being smoked, how much one smokes at one time and on and on. So again AT MOST it will stay 30-45 days. As far as how they test for it, they test for the chemicals themselves and the metabolites of the chemicals. What that means is that when our body processes say, cocaine, it will use up the cocaine and have a remainder that is left to be disposed of through the urine called norcocaine. So if your high while taking the test, the cocaine shows, and the next day, you havent gotten rid of all the metabolites so the norcocaine shows.

why are they testing for nicotine I am uncertain, but if you are only smoking three times in a month stop and your blood should be clean in thirty days. Is nicotine a euphemism for something else, if it is then marijuana clears in thirty days also.

Nicotine is not an illegal drug. Smoking cigarrettes where nicotine is found is not illegal. Now if you were smoking something illegal, they will find that drug, but its not nicotine.
