depends what style and in what key is the song played. there isn't a fixed formula for that, you just need to learn from experience what sounds good.
but as a rule of thumb for standard backup singing, you usually are safe if you sing two notes above or below the lead singer (or can be one or three above or below, depending what chord fits the melody in that measure)
e.g. measure1: C chord
lead singer: c1-e1-g1
backup singer1: g0-c1-e1
backup singer 2: e1-g1-c2

You can just hear when it sounds right. You should have an understanding of chords and harmony to really grasp the concept of singing back up.
