Answer honestly, and try to put a positive spin on it. Also explain what you have done in the past to get through your weakness. For example "I tend to get very emotionally invested in my work, which can be a problem because I lose objectivety and can get a little bit burned out. But I've learned that if I get used to sharing some of the responsibility with my team-mates, it is easier for me to let go when I need to." It's important to show that you recognize your weakness and already know how to handle them!
They're going to find out over time what your weaknesses are so it's no use hiding them. Also avoid bringing up weaknesses like "I tend to be late too often" because that looks really unprofessional and immature.

Depends on the job for which you are being interviewed. Clerical or accounting job, you might say," I have a tendency to be too detail oriented". Turn a negitive into a positive.

you either 1. take one of your weaknessess (not as familiar with Excel as you would like to be) and combine it with what you are doing to change it (so I am taking classess to strengthen it) or
2. you give them a quality (analysis for instance) and say how you see it as a challenge and so you keep at it until you get it which sometimes you get too into detail. Just a suggestion.
