Mother may I call you Sister or Mother Superior, or Sister Superior, just kidding , the correct title would be: Sister Mother Superior!

If you're close to her, you may write "Dear, Sister Mary"...if you are just meeting her, call her with her last name "Dear, Sister Smith"...Our sisters don't mind if it's by first or last name because there is no rule in addressing a person by first or last name in catholic practices

just use the christian name, sister mary or whatever it is. thats a religious name given to the nun when she enters the convent and is used for all formal address. if it was someone you were close to you could use her actual name, my aunt was sr alphonsus but we called her auntie winnie cos that was her actual name. my other aunt was a nun, dont even know what her religious name was cos we only ever called her auntie rea.
