It makes zero difference. THE WAY YOU CHECK THE TRANSMISSION FLUID! The vehicle needs to be running and have run long enough to get the tranny fluid hot(10 minutes or so) cycle the shifter from park to reverse-to drive-and back to park. pull the transmission dip stick-and check the fluid level. The add mark means ONE PINT(NOT ONE QUART). You may add fluid to a running engine or a stopped engine. You should not overfill the transmission.

Add fluid when the vehicle is off. Check it when the van is running. Make sure you add the correct transmission fluid (plus 3) NOT DEXTRON!!!

It's best to check your transmission fluid when your car is warmed up, but you can add it whether it's on or off.
