Manga - Hana Yori Dango 花より男子
Anime - Boys over flower

If you would have asked this question a month ago I would have said ai Yori Aoshi as well but you have to check out both anime (Boys over flower) and manga Hana Yori Dango. Its quite an amazing story.

if u're about high school/college age, u'll love this one: "Love Hina" the comic (manga), not the anime (i was really dissappointed in that). Another good comic is GTO. That one is a bit older but it's probably one of my favorites. The anime for GTO is more or less OK.
For jap animation, my favorite so far is Monster. It's for more mature audience though, at least have to be in college to truely enjoy it. It's grim and ironic, but it's a really well done.

For american comics, I like Calvin & Hobbes, and Dilbert (u need to work in an office to really grasp these jokes though). Animation wise, back in the day my favorites were Inpector gadget and Duck Tales. They probably don't exist anymore though. Well I was a kid then so I can't tell if they would match ur taste anyway.

P.S. Other people mentioned Naruto. Yes it's good too. But read the manga. It's better. The anime is OK up to around episode 125 and then turns to utter crap.

"Zot!" by Scott McCloud
"The Spirit" by Will Eisner (particularly the post-WWII stories)

Best animation:
1940s Fleisher Studios Superman cartoons (still the best!)
Chuck Jones' Looney Toons cartoons (all of 'em!)
