I recommend
1. The supermarkets (absolutely superb in Amsterdam especially the freshly prepared food)
2. Attached to one of the supermarkets there is an open food emporium with all sorts of dishes made there and then for you to eat. Great food.
3. The Van Gogh gallery is worth a visit if you appreciate good art.
4. Anne Franks place is a bit crap, watch the movie instead
5. Try going on a tram trip to the outskirts of Amsterdam, their public transport system is great
6. The canal tours are a novelty item if it is a sunny day.
7. Hire a bike and go slowly as it is dangerous on the bike routes which have kerbs so cars cannot drive there.
8. Tulips outside of Amsterdam. Look out for the black tulips, highly prized.
9. Visit a windmill, and marvel at the foresight of the forefathers who foresaw that one day oil would run out
10. There are some great places outside of Amsterdam worth visiting.

The Van Gough Museum was unbelievable. Also the Heineken Brewery was great - you get a free beer in a souvenir glass at the end of the tour! I also saw Anne Frank's House which was very interesting. Hire a bike and just cruise around the cobbled streets you will come across many more sights and attractions! What a fantastic city. Enjoy!
P.S. But do make sure you check out the "obvious" attractions ;) It is all part of the experience after all!

Wow, that's new! 8)
Hmmmm.... U need a good pair of shoes, a light(!) backpack and you can walk from morning to night through the streets. It's gorgeous! Grab yourself a good travel guide, i think, Rick Steve's books are the best. No I'm NOT advertising for him! 8) U should look some things up on the Internet. There are good music lounges and clubs! U'll have a lot of fun there, mostly friendly people there.

You have all the touristy places listed below but if you want to check out the trendy cool places that the locals go.. here's some tips
Eat Poffertjes (small mini pancakes - to die for) before you leave - make sure you get them fresh.
Dark chocolate. Get them at the Albert Heijn supermarket (cheaper and lots of variety) Good for presents & yourself.
Drink lots of cappuchino - about the same cost of a coffee but infinitely better than in the US.
Heineken brewery tour (for the price of entrance you get 2 beers and a souvenir).
While you're being a tourist, check out Cafe Chez uzzi, near Waterlooplein across from Mr. Visserplein tram stop. Cheap beers, good eats and there's usually a good crowd on Thurs/Friday nights.
Clubs/ Restaurants: Jimmy Woo's (if you can get in), Hotel Arena, lots of bars in de Pijp (near Albert Cuypstraat where there's a huge flea market). Cafe de Jaren (go outside on the terrace if the weather is nice). Panama. Note that cocktails, if you can get them, are pretty expensive & weak; water is not generally free. Plus you have to pay to pee - the toilet ladies are pretty militant in the bars. Beer and wine is cheaper ! Vak Zuid, at the Olympisch Stadium. Cool place for fusion tapas and you have a view on the stadium itself.
Go out to Java Island (in dutch eiland) - away from the tourists and where the locals live. You'll see some of the coolest architecture (if you are into that) and some really nice houses. It's also on the water.
Another interesting museum off the beaten path.. Tropen museum, about all the places the Dutch colonized/enslaved.
BTW, the comment about supermarkets in Holland being superb? Forget it. No place in the world has better supermarkets than in the US of A. But if you like cheese & chocolates & coffee.. load up here!

Boat rides in the canal, flower market, a wealth of museums (as mentioned by other answerers) and Madame Tusseau exhibition. Also, watch out for Boom! Chicago standup comedy shows (in English)
Also, Amsterdam in just a stone throw away from these cool places by train:
-Haarlem (10 mins): beautiful Grote Markt, Amsterdamsepoort, cathedral
-Hoofddorp (10 mins): cosy industrial town near Amsterdam airport
-Volendam (??): Fishers' village/marina, nice walk, fresh fish, and photos in Dutch costume
-The Hague/Den Haag (45 mins): International Court, Queen's Palace, Maurits museum, Tweede Kamer, Scheveningen beach, Madurodam (the entire Netherlands in miniature)
-Utrecht (45 mins): ancient city, riverwalk, fine dining
-Rotterdam (1 hour): modern city, theatre, fine arts, fine dining, harbour
