According to the US Treasury Bureau of Engraving and Printing, the 1862 $2 note had a portrait of Alexander Hamilton. Jefferson's portrait was not used until 1869.

The 1862 $2.00 bill is Friedberg # 41 (FR# 41) It was Design #8 and has Alexander Hamilton on it, facing right. It was signed by Chittenden / Spinner. It is what is called a legal tender note.
