I made a purchase online a few days ago with my Mastercard Debit. However, the transaction got cancelled before it had a chance to charge my checking account. However, my "available balance" on my statement still shows the deduction from the purchase. Should I be concerned?

No...it should fall off after a few days. Places often hold an amount, and once that pending charge falls away you should be fine. If it's still there in a couple of days just call your bank.

If the available balance on your statement still shows the deduction from the purchase, then the transaction did NOT get canceled before it had a chance to charge your checking account. You best check with your bank.

The charge should be removed in about 8 days from the date of cancellation. That is just the authorization merchant used. If it is not removed after 8 days. Dispute the charge.

Yes, if they deducted the amount of the purchase from the vailable credit limit, it means that to them, it is the same as if you got your merchandise. Have the company you made the apttempted purchase, issue you a credit if the purchase was indeed charged to you wrongly.
