This question looks like its been answered pretty well but I wanted to throw in my two cents. Make sure that the folder on the desktop does have an arrow on the bottom before you delete it. Without the arrow you may be looking at the actual file folder. When you delete it will ask "Do you want to delete this short cut?" You're cool. If it asks "Do you want to delete this folder and all its contents?" You're in the hot seat. You'll have to put the folder back where you got it from.

If you add icons to the desktop they are safe to remove because they are only there, so that you can get to your files or connect to the Internet more quickly than searching for the files.
If you click the right side of your mouse on something you be able setup up a icon on your desktop for that file.

BUSTED!!
A) Stop being sneaky.
B) O.K., enough chastising. Here's what you do: (Providing you know the original location of the file.)
Right click the file. A menu will appear, select "Send to", then select from the choices that appear.
C) If you don't know the original location of the file, click Start, then "Help & Support", then click "Undo changes to your computer with System Restore", then click "Restore my computer to an earlier time".
Select the most recent restore point available, then click "restore my computer".
That will put the file back in it's original location, without deleting any recent file work.
Good luck.

Remember to press the "Esc" key if you are unsure about any action.
Please check if it is a real folder or just a shortcut (if it has a small arrow on the left corner- you can delete it).
Do you know the past position of that folder? You can drag him back the same way you brought on the desktop.

If it has a little arrow in the bottom left hand corner, then it is just a shortcut. You can delete it, it will only remove the shortcut from the desktop, and not the file itself.
If it does not have an arrow, then you are stuck I am afraid.
To delete it, click it once, so that it is highlighted, then press delete.
