Go into your START menu and pick Programs.. then Accessories... then Entertainment. Pick the SOUND RECORDER and try recording something from your microphone. You can easily record a few sections, hit STOP then hit PLAY to run a quick test.

Then it is a matter of looking at the plug. Then checking the sound level. Just double click on the Volume Control on the task baclick on RECORDING. Click OK and you will see the Microphone volume control. CRANK IT UP and test using the Sound Recorder again.

Many of the IM tools have test and setup features built in to them. If you can work through those settings and you can hear yourself talking through your mic over your headset or speakers then everyone else should also unless you have such a slow internet connection that a meaningful data stream cannot be maintained.
If you are running Win9X or ME then there may be little hope of you being able to do too many operations at a time without disabling many other programs running in background. Try turning off other hardware and software that is not 100% necessary. Any external drives, Indexing services, Virus scanners, Firewall software, anything and everything that you can except the IM or cam software you are running. If that works then you need an upgrade in hardware (memory & CPU) and software (OS).

There are many ways to check the working condition of your mic but I'm putting here the easiest one...
Goto
~Start Menu
~Programs
~Accessories
~Entertainment
~Sound Recorder

Now click on the "Record Button" (The big red dot) and now start talking in your mic, if you see the graph in the sound recording varying with your sound your mic is OK, and if not then you mic is not working properly.

If your mic is woking properly and still you can't have a voice chat you have to properly configure your mic in your yahoo messenger for this, Goto your messenger and from there
~Actions
~Call setup
Follow the wizard, and your mic will be configured.

(If you are using YM 8 I'd recommend you to switch to YM 7 for best performance.)
