Matrix Assisted Laser Desorption Ionization Time of Flight. MALDI is a method used in mass spectroscopy to gently fragment large bio-molecules like proteins. The sample molecules are placed in a matrix that can be excited by a laser pulse. Energy is transferred from the matrix to the sample molecules causing fragmentation. A second laser desorbs into a gaseous state sample fragments and a portion of the matrix. This mixture is fed into a time of flight mass spectrometer for analysis. The original sample structure can be inferred from the fragments that are produced.
