It's under the State Dept. Ambassadors are selected by the President and approved by the Senate. The Senate thinks it fun every now and then to hold up Ambassadorial appointments in order to get the President to support some bill or other. Jesse Helms used this tactic often on Clinton, and even some on Reagan.

It depends on what you are referring to as diplomatic service. I think the State Department and through its embassies establish the formal lines of communications with foreign countries. However, diplomatic functions are often carried out through various other channels as well such as the NSA (National Security ), Commerce department and etc depending what the issues are, sometimes openly and sometimes quietly behind the seen. President may also chose to send different cabinet members on diplomatic missions based on his judgment as to who is the right person for that particular issue.

Others have answered and they are correct in saying it is the State Department. All persons selected for these jobs are via "appointments" by the Executive Branch - the president, et al.
