I guess the first thing you should do ..even before you say something to him is figure out how that made YOU feel. Ask yourself...What does it mean to me to have a brother that might be gay? If you realize that you still love him no matter what then it doesnt matter.
If you realize that what you think is ..."****, that is sick" or "I dont want a gay brother", then I would suggest trying to find a way for you to deal with it.....
All in all ask yourself Why do you feel like you have to do anything at all?

Since he is only 16. You should inquire as to what he was doing? Playing games or was he in a serious chat. If it was the latter, then you should discuss with him the dangers of using the internet for such purposes. As far as the gay issue, so what. Whatever drives your bus.

observe him and see if he is sexually confused if he is than talk to him but if he's confident than leave him alone cz its his life you shouldnt oblige him to be straight
and maybe he was just looking for smth new and just trying it or maybe he's making fun of those who are chatting
