If they are Syrian hamsters, don't even try it.
If they are dwarves, you can occasionally introduce two and have them cohabitate with no problem. Other times, though, they will fight anyway.
Dwarves are social creatures, Syrian's are solitary creatures. Dwarves do get territorial, though, especially if they have been alone for more than a month or so. If your hamster has been on his own for a long time, it's probably not wise to introduce them to one another, as the older one will fight with the younger one.
Your best bet on trying them together is to use a large rubbermaid type storage tub, deep enough that they can't get out, and put in clean bedding. Then, add a small bit of bedding from the older hamster to 1 corner, and a small bit of bedding from the younger hamster to another corner. Add several hiding places, such as little houses, but not tubes (if they fight inside the tube, it's hard to get to them to seperate them. Coffee mugs turned on thier sides work great.
Next, introduce the younger hamster first, let him run around the tub a bit, then put the older one in. Please note- do NOT attempt this with Syrian/Golden/Teddybear hamsters!! Do NOT attempt it with mixed sex pairs (male/female).
Wear thick gloves, and sit right there. They'll eventually start sniffing at each other. As soon as they do, they'll either ignore one another, sniff one another more, or one will attack the other viciously. Seperate them IMMEDIATLY if that happens.
If they ignore one another, or just keep sniffing one another, keep watching. Just stay right there next to it, to make sure that they don't fight.
As long as they don't fight, keep them in there for about 30 minutes together. Do that 3 times a day for 2 days. As long they continue to get along, and don't fight viciously (they might start to playfully wrestle- you'll know the difference when you see it), then you can add them in to a cage together.
Make sure the cage is large enough for two hamsters. A basic, slap together crittertrail cage is NOT big enough for two full grown adult hamsters, even dwarves. Add on to it (if this is the type you have) with tubes, little houses, or even another cage.
Make sure it has been completly cleaned out, soap and water, scrub, scrub, scrub. You'll pretty much want to wipe out the scent of the older hamster, so that they have thier own territories.
New bedding is needed, but also do the same trick as in the tub- add a small bit of their own bedding to two seperate sections in the cage, usually each with it's own hiding spot like a little house.
Only use carefresh or Aspen bedding, never PINE or CEDAR, as those are harmful to hamster's lungs.
Also... for all the people who are constantly remarking that ALL hamsters are solitary, and cannibalistic, please get your facts right before making a blanket statement like that. Dwarves are the exact opposite- they are social creatures, and want companions!

Hamsters are territorial. The big one will pick on and maybe kill the little one. The only time you can keep hamster together is if they are littler mates. Then the chances of them fighting as they get older are high. Not a good idea to house them together.
