I saw a documentary on this once. There was an ancient civilization in Illinois called Cahokia that became a very large city for it's time. The people abanoded it before Europeans arrived to the area, so little is known about them.

Depends what you mean by large. And are you including the central and south american tribes? As far as North America goes probably the Mounds Indians left the largest earth works.
