Even though the play is called in the huddle, sometimes when the quarterback sees something he doesn't like in the defence, he'll change the play at the line of scrimage. Those numbers and colors correspond from a secret code that only the offencive players know.

All of the above are correct but usually its mostly play calls or tricks to making the defense think its a play call. They are all codes that should be understood but only members of the offense or members of the team calling the play. Also it could be a way for the QB to let his blockers or Ends know who to watch out for on the opposing defense.

Sometimes the color is the formation and the number is the specific play. Other times each is used to decoy the defense. The offensive players know which is which. The defense does not.

Play changes, route changes, blocking calls..formations...it's all part of the things the QB has to do before the play to get the best outcome, sometimes it works, sometimes it doesn't.
Ah also...he'll try to draw the DL across to get a flag too..you'll see that a lot.

Sometimes it can mean certain coverages or defensive alignments. "Red" might mean "on the right side", while "blue" might mean "left".
Sometimes it doesn't mean anything and can be meant only to try to trick the defense into making some adjustment.
