Yes, it's possible, with some caveats.

* The disk must be formatted as FAT32. NTFS won't work.
* It won't replace the Xbox 360's hard drive
* It will only allow you to read data off of the disk, not write data back to the disk.

In essence, the hard drive is acting like any other USB mass storage peripheral you might attach to the Xbox 360 (iPod, flash drive, etc).
