With the engine and exhaust system cold, raise the hood, then start the engine. Quickly, while the parts are still cool, feel around the exhaust manifold with your hand to feel for the telltale puffs of air from a leak. You will have only a few seconds before things warm up, so work quickly.

Although your missing a stud the manifold gasket should protect the surfaces for a time. heat won't close the gap You need to carefully drill out the piece. start with a small drill then get bigger till it comes out,or drill a hole and use a stud extractor, run a tap down it and replace the stud. You may as well remove that manifold first as it's bound to need a new gasket. It will make access easier too.

you need a helper.get the toyota up to operating temp. have your helper wear heavy gloves and hold a handful of rags at the tailpipe to create excessive back-pressure in your exhaust system.while they do this you can check around in the engine compartment and underneath the vehical.dont plug the exhaust more than 1-2 min.at a time or you may blow other gaskets.good luck.

You can also use a piece of fuel hose up to your ear and check around the exhaust system. You will be able to hear a leak.

OK, this makes a mess but works.
Mix some flour a water, so it kind of gummy.
Paste it on the exhaust, and crank it up.
The leak will blow a hole in the slimy mix.
Hose it off when done

I use a piece of 3/16" brake line about 18 inches long with a length of rubber tubing on it. then I can put the tubing to my ear, and I can bend the tubing to go around the manifold and reach places I can't reach with a stethoscope.

Soapy water. Spray it around the area where you think it is leaking. Mind you do this when the engine is cold.
