I'm assuming you mean the region of "Babylonia", not the city of Babylon itself. In fact, you likely mean the region of southern Mesopotamia --the area between the Tigris and Euphrates rivers down to the Persian Gulf. (It was only called Babylonia once Babylon was established as its capital, long after the first cities appeared.)
In that case, the most ancient cities are those of the Sumerians. They build a cluster of citiy-states which became very prominent in the early 3rd millennium B.C. Perhaps the earliest of these was Eridu, very near the Persian Gulf. Another very important early urban center was Uruk (biblical Erech, modern Warka). Urbanization of this region (something more than just 'the earliest settlements') took place during the "Uruk" (named for the city) and "Jemdet Nasr" periods of the 4th millennium B.C.
Whether one chooses Eridu or Uruk as "the first city" depends in part on when you consider a settlement "urbanized".

babylon is a city, it is the ancient city which would be in current day Iraq. it was renowned for it's hanging gardens (one of the seven wonders of the world was the hanging gardens of babylon) it is also famous because it is were the infamous tower of babel was. the story is that the people who built the tower were so proud that they believed the tower would reach the heavens. to punish them the gods made it so that they all spoke in different tounges and couldn't communicate to finish the tower (orgin of the word babble) This story was meant to explain why different languages exist
