There's a bolt that holds the tire in place. Remove that, measure your tire, and go to HomeDepot or Lowes (Or a DIY Store) and buy a new tire.

More information would be helpful, but do you mean actually replacing the tire or just getting air back in it? Generally, a wheelbarrow's tire is tubeless and can be cantankerous to get a seal on both sides of the rim to allow inflating. A good bungee chord wrapped around the perimeter is usually enough to get a seal. It's sometimes easier though to just get a tube, cut off the old valve stem and install the new tube/tire.

take out the wheel stud take the tire w/you to a building supply store to buy a new one
