Moisture in the room possibly and the fibre the rug is made of can possibly be an attractant for mold.

There is dampness coming from somewhere. Mold likes it wet. Have you checked under the rug?????

MOISTURE1 JUST BECAUSE THE CARPET IN NOT WET DOES NOT MEAN THERE ISN'T MOISTURE. IF THERE IS WATER UNDER YOUR HOUSE THE MOISTURE MAY BE COMING FROM UNDER YOUR FLOOR BOARDS. IT DOSE NT TAKE A LOT OF DAMPNESS TO FEED MOLDS. AT SOME POINT YOUR CARPET GOT WET OR YOUR FLOOR GOT WET AND THE MOLD SPORES ARE ALREADY THERE YOU NEED TO HAVE YOUR CARPETS CLEANED WITH A MOLD KILLING SOLUTION OR REMOVE THE CARPET, AND USE A BLEACH ON YOUR FLOOR. ONCE MOLD IS IN YOUR HOUSE IT IS REALLY HARD TO GET RID OF.

Water causes it.. and it lives very well in damp places with paper.. like drywall. If it is on the walls, they have drywall now that is paperless... good for damp areas. I hate mold.. some can be very dangerous!! Since it is on the rug.. do you have hardwood floors? Check your crawl space for standing water.. your floors may be taking a hit if the crawl space is wet.

Possibilities are innumerous, especially since mold spores are disbursed through the air.
but if its a nice rug you dont want to chance ruining with detergents etc you can try using ahtlete foot powder sprinkled over it.
