Reading comprehension, as it is, is reading a given passage, and it test your level of understanding to it.

The sentences in the passages will tell you something directly or indirectly. So, you should be able to infer and predict. Infer is like to assume what's going on. For instance, if the passage described a boy standing at the window of a confectionery shop, staring at a cheesecake with tears in his eyes...., depending on the rest of the passage, it may means that he wants to buy one but he could not afford, or it may remind him of a past event.

You are supposed to display as much as possible that you understand what the passage is trying to tell you, that is where you gain points. But you should also read the question and make sure you understand what it is trying to ask.

Reading comprehension is a part of many standard exams like GMAT and TOEFL. Get a book or CD on either one of those exams and you will find a lot of practice materials there.
