Herbarium is absolutely right. Black Widows to not congregate in large populations and they are reclusive enough I really doubt that is what you are seeing in your back yard. Their webs don't look like a regular spider web and are almost always in dark, out of the way locations. Double check and make sure that is what they are before you start an annihilation campaign. The Common Black Garden Spider is often confused with a Black Widow, even to red markings on the body in some varieties, and are very beneficial.

if you "kill" it I'd say it's dead then.
if you spray it directly with ant spray it may kill it
but then again if you spray it with ant spray and it scampers away i would have to say no it's not dead just relocated.
spiders walk on their tippytoes so if you spray poisons for them unless they are directly sprayed that wont kill them, unlike roaches and ants that are constantly cleaning their feet and antennas. that's how the sprays kill them. they get the poison on
their feet or antenna and clean it off with their mouth and get the poison in them that way and die. spiders on the other hand don't clean their feet that way so.... go figure

THe folks at the university extension had said the best way to get rid of spiders is to vaccuum them up. that way they go through all that tubing and end up in a bag and really don't have the brains to get out of there.but if your squeamish you could remove the bag and thow it in the trash and viola! no more spidey!
