Virginity is defined in many ways. There is physical virginity, which means that the female's hymen, a fine membrane that is found at the opening of the vagina is intact. In this day and age, a young girl often looses that "physical virginity" with her first trip to a gynecologist. When a speculum is used to examine the vagina, the hymen is usually ruptured.
Then there is the person that has not ever had intercourse, this is another form of virginity. So, no, lack of blood on penetration doesn't necessarily mean the female has had sex before, it simply means her hymen was ruptured earlier.
