Your best option is to go directly to the hotels website. Most of the major hotels in vegas offer great packages including air and rental car. If you have a hotel in mind you should go to the website and see what they have to offer.
If you are totaly undecided.. go to an airlines website.. such as northwest.com or whatever airline you fly the most. They also will offer packages.
Going to a website like expedia or orbitz may offer you alot more upfront choices, but the prices are likely to be a little higher.

Expedia.com or hotels.com orbitz.com or try one of the actual names of the many casinos and hotels in Vegas and some times they have packages. Shoot here where I am they even fly a plane up from Reno that one of the hotel/casinos owns and people purchase the airline 2 way ticket and a 2 night stay for the resort so you can check on that stuff too. They want the business so why not fly their private air lines?

I just booked a 4 day trip on orbitz. airfare+Mandalay bay+ rental car for 563 a person! That was the best deal I could find. I would recommend ORBITZ!
