It was in return of the Jedia, towards the end when Luke was fighting him. He sensed it from Luke in the fact that Luke has a sister and hence, he was her father.

Darth Vader never actually found out that Princess Leia was his daughter, just that Luke had a sister and we are lead to believe by George Lucas that in the same film, 'Return Of The Jedi', when Luke tells Leia that they are brother and sister that Darth Vader also reads that in Luke's mind while they are fighting.

Well Anakin was told by the emperor that the girl had died, after he'd assaulted her and probably assumed the babies died with her. But I guess as soon as he found out there was someone else with the surname Skywalker who could use the force, he was smart enough to work out he was the Dad, so I'd say The Empire Strikes Back, since he doesn't really meet or pay attention to Luke in Star Wars. He certainly makes no sign of realising Leia is his daughter either in Star Wars. I can't remember if he knew there were twins, but if he had access to her date of birth, I would have thought he'd have worked it out before reading Luke's mind in the third film, since he knew already the force was strong in her.

I believe it was episode 6.. Return of the Jedi.. He had already told Luke that he was his father.. later on, Luke's thoughts betray him and Darth Vader sees the thoughts.. he can DO that, you know.
and for those who keep saying that Darth Vader always knew.. NOPE.. he always knew he was LUKE'S father.. but he never knew about Leia..

Return of the Jedi - Luke 'leaked' it telepathically when they were fighting at the end of the movie.
Darth wanted Leia killed at the start of the 'first/fourth' movie "A New Hope" without knowing who she really was.
But the name actually translates as 'Dark Father' - so it should have been pretty obvious to everyone!

Darth Vader always knew deep in his soul which is the reason he never killed her or put his hands on her.
