It is not intended to be in any real state. In one episode they say that the Simpsons are from North Kentucky, but the creator Matt Groening said that it is similar to his home in Portland, Oregon.

Classic question.
I think its an east coast, mid-atlantic state since Homer used New York City as a hub for his bus trip to go to the Burlington Coat Factory in Pennsylvania. This is something only someone south of Massachussets and North of Virginia would do.
Of course that doesn't explain that Springfield gorge looks western, and the house has mission style round top doors, but it takes care of everything else.

According to the episode, 'Behind the Laughter', they live in North Kentucky. But it's interesting to note that only two states in the U.S. don't have a town called, 'Springfield'.

I don't think anyone knows..they only mention the city of Springfield which is a common city name all over the country. Do you remember when they were on the Tracy Allman show years ago and were green? That's where they got their start.
