There are no nonstop transpacific flights from DEN, so you would have to make a connection. My guess is that you would connect in Seattle, Portland, San Francisco or Los Angeles, or maybe Chicago, Minneapolis or Vancouver.
A flight from the US to Japan takes 10 to 14 hours depending on where you depart from. If you count your flight to the connecting city and your wait time there, your total travel time would probably be 16-19 hours, maybe a bit longer depending on the exact schedule.
If there was a direct flight, I would estimate it at 12-13 hours, because DFW to Tokyo is 13.5 hours and passes right over Denver.

It really fluctuates according to the winds. So from Seattle, you can get to Tokyo in about 9 hours. From Denver, it would be more like 13-15hours. With strong winds it could be up to 17-18. So bring lots of stuff to do.

From Denver, from fact I know it takes 18 hours from Florida to Egypt so from Denver to Japan, maybe longer.
