first, determine if they are casting right now
if so, contact an agent that is familiar with the show/process

Get yourself an agent, work on your standup comedy skills, do as many gigs as you can. Build a reputation for yourself, stay off Yahoo! Answers, keep yourself busy and completely dug deep in comedy. Score auditions for small roles in low-budget movies, work some more on your standup routines and become a regular at the comedy clubs in your neighbourhood.
Find a photographer, and take headshots. Make yourself a resume. Then have that agent of yours try to score you an audition with the MadTV cast.
Hopefully, it'll all work out as planned. If it doesn't....then you really must not be funny, cuz they've had their share of talentless acts on the show!
