If you have actually removed the correct screw, (the one on the edge of the door, inline with the cylinder), then the reason it will not come out, is because the cam in the middle of the cyinder is slightly offset to one side and therefore catches on the inside of the lockcase, the only ways to get round this, is to:-
1. Turn the key slightly to either side whilst pulling on the cylinder.
2. Turn the thumbturn (if you have one on the cylinder) to either side whilst pulling on the cylinder.
3. Drill out the cylinder, turn the cylinder plug (where the key goes in) with a screwdriver either direction, again whilst pulling the cylinder out of the door!

Hope this helps, good luck! ;-)

If you are referring to a commercial door, the cylinder and thumb turn are held into place with a small slotted set screw in the latch mechanism. Remove the set screws and the cylinder and thumb turn will unscrew from the latch mechanism in a counter clockwise direction.

You have missed a set screw. Remove the "Throw" trim on the door edge. Behind that trim you will see a small slotted screw loosen that screw and the "Cylinder of the lock will unscrew.
