-In the 1949 episode Bad Ol Putty Tat - Sylvester lures him with a fake female canary(that he painted on his fingers)
-Granny always addresses her pets (Sylvester, Hector & Tweety) as boys
-Male canaries sing & females do not sing - so he sings therefore he is a male.

I think it's hard to tell with birds, because the male organs stay inside until needed.
