Hiccups are caused by spasming of the diaphragm, the muscles under our lungs that controls our breathing. Just like any muscle spasm it's best controlled by slow, even stretching of the muscle. Any hiccup cure that can do that, should work. Drinking water upside down is one of the the methods that attempt to relax or stimulate the diaphragm which tend to be the most effective.
If you have chronic hiccups, drinking a glass of water upside down does not work. But scientists at the Walter Reed Army Medical Center found that nifedipine, commonly used to treat migraine headaches and to control high blood pressure, cured five of seven chronic hiccup sufferers they studied. Nifedipine keeps calcium from being absorbed by certain brain tissue, Dr. Jabbari said. In some people, inappropriately high calcium levels in the brain may trigger the onset of hiccups.

It's not the drinking, it's the bending over. The glass just helps you to bend over far enough, because in order to drink the water you HAVE to get your head down there! It compresses the diaphragm - hiccups are caused by spasms of this muscle, and compressing it by bending over to drink helps stop the spasms. Holding your breath is the same sort of thing, as is the upside-down spoonful of sugar, I imagine, although I haven't heard that one before!

I have one way that always works for me. I drink a class of water for 10 seconds without stopping for a breath. Then I usually burp and no more hiccups. Hope this helps! =)

Home remedies which interrupt or override the spasmodic nerve circuitry are often effective. Such remedies include:
holding one's breath for as long as possible
breathing into a paper bag
swallowing a spoonful of sugar
bending forward from the waist and drinking water from the wrong side of a glass

Well if you believe in something, that helps it work. I think it got started in the old movie "The Philadelphia Story" where Cary Grant shows Katherine Hepburn how to do it to stop her hiccups.
