I would have to say it is from the movie "Highlander" When Ramirez (Sean Connery) is telling MacLeod (Christopher Lambert) about the "quickening".

Ramirez: Why does the sun come up? Or are the stars just pin holes in the curtain of night, who knows? What I do know is that because you were born different, men will fear you... try to drive you away like the people of your village.
[MacLeod turns his back]
Ramirez: You must learn to conceal your special gift and harness it until the time of the gathering.

Although all of Sean Connery's movies are great. I remember the very first movie I ever saw with him in it. It was "Darby O'Gil and the Little People." I was very little and it was playing on "ABC Disney Sunday". I was hooked on Sean Connery ever since.

Best Thomas "Sean" Connery moment has to be the Indiana Jones and the last crusade scene where he's trying to get the lighter to burn the rope to free Indy and himself.
"the floor on fire.....and the chair!"
"Indiana? we called the dog Indiana."
class act all the way a truly brilliant British actor
Bond? He's the only Bond for me.
Time bandits great, he even made Prince of thieves bearable.
League of gents "Eyes open boy I can't help you all the time"
again showing that father like role from Indy.
And of coarse the Rock the only film that Nicholas Cage doesn't seem boring and drab.

I have to say "The Rock"... what a great Connery movie that was.

Prior to that, who can forget Sir Sean in "Darby O'Gil an the little People", and seeing Sir Sean sing lol.

Playing Dr. Henry Jones Snr. in Indiana Jones and The Last Crusade. He shows that not only can he play the serious father but also still be a playful school boy at the same time.

In the movie "Playing by Heart" he is confronted by his wife played by Gena Rowlands about an affair she thinks he might have had with his secretary years ago, which he tells her he did not have (and sorry, I don't know the quotes word for word, but) Gena says 'so you didn't want to sleep with her?' and Sean says 'Oh God, yes I did!'

"The Untouchables" was without a doubt one of his greatest movies. He is the typical "old school" cop. Awesome performance throughout the movie.
"Finding Forrester" was different, but very good.
However, his "perfect" role was in "First Knight" (he makes a good king and the visual impact was fantastic).

It's from my favorite Sean Connery movie, "Marnie". He shouted that line to Tippi Hedrin in an attempt to cure her of schzophrenia induced kleptomania. Awesome movie. Those who haven't seen it should to so. Connery is in top form and gives another Oscar worthy performance.
