Yes they can, If they both have an ethernet port, all you need to do is connect them with a crossover ethernet cable. This is different than a regular ethernet cable because as its name implies, it has some of the cables crossed over. You'll need this if you don't want to use a router. Once you have the cable do the following:
1) Assign each PC a static IP address such as 192.168.1.10 and 192.168.1.20
2) Assign them the same subnet 255.255.255.0
3) Make sure that you make them members of the same workgroup (not domain). Example: Home or Office
Although most times the computers will them find themselves after these steps, you may need to reboot them in order for the new network settings to take effect. However, keep in mind that if one of them is currently connected to the internet via the ethernet port, you won't be able to use nor share the internet (without a router) unless you have a second ethernet port on one of the machines.

If you have Ethernet (LAN) card on both computers:
connect both computers with a straight through (regular Ethernet/LAN) cable and configure both computers to use static IP address
1st computer:
IP: 192.168.1.2
Sub net Mask: 255.255.255.0
Default Gateway: 192.169.1.2
2nd computer
IP: 192.168.1.3
Sub net Mask: 255.255.255.0
Default Gateway: 192.169.1.3
If that doesn't work, keep the the same IP settings and try a Crossover network cable

There are several ways:
1.USB-USB Cable. Use 'special' cable for this purpose.Don't connect normal usb-usb cable, though.
2.Laplink cable. See different kinds of cable.chepeast is parallel laplink cable.
3. Two USB Wifi. Connect through AD-HOC.
4. Lan Cable. Use cross-over cable if you don't have hub.
