No, not really. If you NEVER ejaculate, you can have a higher risk of some sort of cancer than someone who does (or so I've read), but you can fix that problem by masturbating occasionally. The theory is that ejaculation cleanses your body of some toxins that build up between ejaculations. I don't think you need to do it more than once per week to cleanse your body though, and you might be doing just as well to do it less. Most men masturbate more than that though.

Your physical body, no, other than blue balls.
Emotionally and mentally, yes it will affect you. It can make you form beliefs or images about yourself that are not true. These can become so entrenched that they affect you life, and in turn affect you physically. Just like the effects of marriage cause a lot of men to gain weight. The body follows the mind.

Well, what do you call "regular"? No matter how you define "regular" (once a day/week/fortnight/month/year -- whatever) there are a whole bunch of guys who would fall below "normal" and they are okay physically.
Depending on your desire for sex and you are unsuccessful at suppressing your urges and wants, you can be affected psychologically by a lack of sex. With enough psychological distress (depression, et. al.), yes one can also be affected physically. One who takes a vow of celibacy (such as a priest) has to be able to suppress the natural urges for sexual intimacy. It is obviously very difficult, but it can be done.
