How do you tune a guitar with a floating tremolo system?

I have a floating tremolo and all of a sudden it just wasn't tuned anymore. one string is off and i just don't know how to go b ack to standard tuning. please help me!

Turn off the tremelo and tune the strings E A D G B E ( That's Fat string to Skinny, or String #6 to String # 1)
