I know you wanted a simple answer, but it depends. In many cases USPS will be the cheapest, but in some cases you will do better with FedEx or UPS because of speed or cost issus.

USPS' express mail service for a flat rate envelope costs $14.40 takes between 1 and 2 days depending on the origin/destination in the US. If overnight matters and you are in the 2-day zone USPS might not work for you.

Additionally for FedEx and UPS, the price varies on the zip code of origin / destination (and weight but you are indicating you are sending a letter).

So my advice is to go to the three websites and enter your specific details. Having done it myself, it takes about 3-5 minutes per website the first time and you can do it faster on subsequent occasions.

USPS is slightly less expensive than FEDEX or UPS. If you are getting a volume rate from FEDEX, they might be cheaper.
