Do you mean "check up" or "Tune up"? If you are just doing some routine "checkup" things, I'd check the oil level, check the air filter, check the wipers for cracks or hardness, check all other fluids (transmission fluid, washer fluid, antifreeze, brake fluid, power steering fluid) and make sure they are at the proper levels, inspect belts, hoses and spark plug wires for signs of cracking, deterioration, or age, check the tire pressure, & inspect the tires for uneven or irregular wear. Pull the wheels one at a time and inspect the condition of the brakes and rotors. You could also pull a spark plug at random to see if it is in good condition and not fouled. In addition, look for leaking fluids, both under the hood and under the vehicle.
If you want to get really serious, do a "mini-tuneup"... oil and filter change, new air filter, new spark plugs, and perhaps even a transmission fluid and filter change if needed. I'd also run a bottle of fuel injector cleaner through your next tank of gas to ensure that the fuel delivery system stays clean ( I do this an average of once a month or so).

Get your owners manual out and read it, for starters. Check all fluid levels, air cleaner and tire pressure. When were your spark plugs, fuel filter and oxygen sensor last changed? Are your tires and brakes good? Need to pull a wheel to check the condition of the linings. Are you changing oil at recommended intervals?

check radiator for antifreeze and thermostat opening when warmed up, check oil should not be black, check tranny fluid at proper level should be red in color, not brown, brake fluid level, check belts, should be no cracks, check hoses, no cracks, hoses should be firm and no bulges,check spark plugs and wires, valve cover gaskets for leaks, check undercarriage for visible leaks on frame, etc.
