1. Change your Browser
From experience, changing your browser (Internet Explorer) to Firefox (www.getfirefox.com) would help a great deal, when browsing the internet, has a great popup blocker, and if you install AdBlock and Filter G for firefox it will block 99% of adverts / popups with adverts you'll come accross.
2. Install a good spyware/adware/malware scanner / protector
WebRoot SpySweeper is a very good spyware / adware / malware blocker / cleaner, it actively blocks them, and can scan your system for them.
3. Install a good Anti-Virus program
Kaspersky is a really good AV as it actively protects your system and scans your email inbox, updates regularly (almost every 3hrs) so its very up to date.

Everyone else is offering good advice. Clean out spyware regularly with AdAware and Spybot (download both from www.pcworld.com, the download page). Webroot is also good. But bear in mind that no program will catch everything. Use all three.
For anti-virus, I use AntiVir, which is free and one of the best rated worldwide (www.free-av.com).
Personally, I use the Google toolbar for pop-up blocking. (Go to www.google.com and click the "more' link above the search textbox, then click "even more" in the pop-up menu; the toolbar link is under the "Search" column; download and install it.), although the Yahoo bar is also good.
There are, however, some sites where I want to see some of the pop-ups so I also use a free program called POW. This is available from www.analogx.com.

Go to the following websites and download the products and updates:
Spybot Search and Destroy 1.4
http://www.safer-networking.org
Immunize then do the scan.
Ad-AwareSE Personal 1.6
http://www.lavasoft.com
Do the scan and remove offending entries.
Avast Home 4.6
http://www.avast.com
If you are running Norton Antivirus or any other AV, DO NOT install Avast as there will be a conflict amongst Norton and Avast.
Always keep all of your products, antivirus and spyware, updated

Well, i have installed the google bar. And it comes with a pop-up blocker that blocks the majority of pop-ups. I would also recommend watching what sites you go on. There's been a deal with IE, and it's weak points (if that's what browser you use). If it is, download IE 7.

well thats a common problem..what i recommend is: swicth to firefox if you are using IE thats will stop most of popups,also get no-script plugin for firefox that will stop most of the nasty scipts from running.Scan your PC with any good antispyware programs like web root spy sweeper/spybot & distroy,Ad-aware..etc then install a good firewall & AV am using sygate with kaspersky thats works gr8 for me.I havent face any spyware problem for last 10 months

Check for spyware. Run both Ad-aware (http://www.lavasoftusa.com/software/adaware/) and Skybot (http://www.safer-networking.org/) to remove any spyware. Run both as one may caught something the other missing. Then go to microsoft.com and download Defender to protect against future infections.
