There may be more than one shut off valve. Check along the line to the hot water heater for any other valves. The line also could possibly be clogged, although that is unlikely. Or you may have a bad valve somewhere, shut-off or sink. Shut off the water when working on any valves so you don't get burnt or have water spraying all over the place.

Unless you are well versed in plumbing you might want to call a plumber. Although you could do it yourself. Simply turn off the water at the main shut off valve and replace the knob or try to rid it of any objects, (such as rust, minerals or whatever) That may be causing the problem.

Don't know what kind of faucet you have. Could be a bit of rust or mineral deposits clogging the cartridge in your faucet if it's a more modern type. Or just a cartridge that needs replacing. Could be the washer became damaged and stuck somewhere it cuts off the flow on an older type. Is the handle stripped and turning when the stem isn't? Could be pipes filled with mineral deposits to the point where they need replacing (rare, but possible).

Shut off Valves do go bad and do not open and close. Be careful. Make sure the water is turned off downstairs before checking if it is the valve.

you have something in the water line in that sink on the hot side. Either unscrew the spout make sure there is not something blocking it or call a plumber

You should have two valves one for hot one for cold check both. You might also check Hot Water heater. Is it on? What are settings?

Most likely some sort of obstruction in the hot side of your faucet. Take it apart and clean it out.
