Ultimate Spider-Man is about 16-17. 616 Spider-Man is about 29 (he's been Spidey about fifteen years and was bitten around age 15).

He's supposed to be about 15 and a sophomore in high school. If you notice, in one of the earlier issues he's stressing out about taking the PSATs, which are usually taken as a sophomore or a junior.
