You really can't stop this activity, you can change to a new e-mail if it gets that bad. I just block the whole domain. I still get a few that trickle in to my inbox which there not supposed to be able to get through to, most junk goes to my bulk mail--which I check for any important stuff and then delete. I would e-mail whoever your Internet ISP is (yahoo, hotmail, aol, etc.) and alert them to the fact that you are getting this stuff and don't want to, maybe there is something that they can do.

If you don't use spam blocking that is very exclusive (such as AOL with a whitelist that only allows certain email addresses, or a challenge response approach), spam filters aren't 100% effective. However, using a spam filter does cut down the amount a lot. I personally use spam filter software called SpamBully for Outlook/Outlook Express where it learns what is/is not spam buy you "training" the software from emails you receive. Even with SpamBully, I still get some emails that come through easily as spam to the human eye. However, the total amount of spam is decreased a lot.

The way that scammers find your email address is by using search spiders to crawl the internet looking for addresses. Have you ever posted your email address onto a guestbook? That's one of their sources. Create a new email address and only give the address to people you know and trust.
Ozauary suggested baiting the scammers - www.scambaits.com is all about baiting and even has a mentor program to teach you how to do it in a safe way.

Report them as spam and they are blocked from your mailbox. Should go straight to your bulk email.

Have you ever thought of scam baiting? If you don't choose to participate, it is gratifying to know someone is doing something to get back at these scammers.
