There really isn't a "for sure" answer on this...
Alot of guys I know are just comfortable around other guys, and some of the things they do to eachother, some might interpret as "gay flirting"... but it isn't... they're just being guys... lol.
Now, on the other hand, I have been hit on by a few gay guys in the past. Usually, unless they're the rare gay type who are just after sex, the gay guy will just be really sweet to you. They'll be interested in what you are saying, make eye contact, and smile alot when they're talking to you. You can tell if they're gay/flirting or not by how they act in a converstaion. Straight guys probably aren't going to be as attentitive or sensitive as gay guys.
Its usually pretty easy to tell... but if you're really close friends, and are still unsure, just ask him! Not in a large group of people, obviously, but when you two are alone. Just ask!

Or when they flirt with you in a obvious way. Sometimes you can't get one to admit it, but watch there actions. This one guy I knew claimed he was straight had a girlfriend, talked trash about gays, the whole thing. But would always sexually flirt with guys. Since the guys he would flirt with were straight and new nothing about the logic of sexual orientation, they didn't really know what his actions meant at first.
