First of all which mail you are talking about.
Yahoo traditional mail won't delete all the mails at once.

In Yahoo beta, you can delete all the mails.

Yes, Select all the emails by pressing control-A (If using email clients) or by clicking a checkbox by the email for web based emails and select delete.

You can also create a rule for certain emails that you want deleted without having to delete them yourself (Email clients only)

just click on the box next to sender, all your e-mails will be highlighted & press delete. all items deleted will be moved to the trash bin so be sure to empty your trash bin as well.
