I listened to a podcast from the producers Damon Lindlof and Carlton Cuse and according to them, the numbers hold no real significance and that if you are looking for some answers regarding the show you should research more the spiritual/moral/philosophic aspect of it.
Now I'm sure that they wouldn't put so much emphasis on the numbers if this where entirely true so I'm guessing they have at least some level of significance. Probably, my best guess would be they are simply the coordinates of the island or the hatch. And all the other stuff like the curses and everything else is just plain weird freakish coincidence.

Each number has different significance. I don't know exactly, and I can't wait to find out more about the Others....
I haven't done the LOST experience, so maybe there are some answers on that web game.

That's the premise of the show.
It'd be kind of like asking, "So where is Gilligan's Island, and why doesn't anyone rescue them?"
The numbers are one of the mysteries of the show and will be explained in due course.
